Blood Sampling

The purpose of the study is to determine the frequency of allelic variants in TPMT in the Hispanic population and to compare the frequency in the Hispanic population with the Caucasian Population.

Relatively few studies have been performed evaluating the variant allele frequencies in Spanish populations. To date, no studies evaluating the TPMT variant allele frequency in the Hispanic population have been reported. Hispanics are a growing part of the patient population treated in the United States. Characterization of the proportion of patients with variant genotypes in this population is important since Hispanic children represent a significant proportion of children being treated with thiopurines.

A single 3 cc heparinized blood sample will be obtained for TPMT genotyping. The sample will be obtained either simultaneously with a clinical sample, at the time of an otherwise medically indicated intravenous catheter placement or scavenged from excess blood remaining following clinical laboratory testing.

Community representatives will be asked to participate in either 1:1 semi-structured interviews with a research staff member or to engage in focus group discussions with other community members. These interviews and discussions will gather broad perspectives on resource and training needs for community lay navigators and inform the adaptation of existing Peer Connect training materials

Lay navigators identified from among the recruited community representatives will be trained either in-person or virtually by research staff members in motivational interviewing and other strategies to provide psychosocial support to older socially disadvantaged adults with cancer. The purpose of this study is to convene a group of diverse North Carolina-based community representatives (patients, caregivers, leaders of faith-based organizations, social workers, and patient navigators) to inform the adaptation of materials used to train lay navigators in the delivery of psychosocial support interventions tailored to older socially disadvantaged adults with cancer.

This behavioral study will explore community representatives' perspectives on the resources and training needed to support the development of a lay navigation program tailored to older socially disadvantaged adults with cancer.

It was hypothesized that diverse community representatives could inform the adaptation of training materials (Peer Connect) and subsequent training of community lay navigators, who will then be positioned to address the psychosocial needs of older adults with cancer

Adapting Peer Connect to Train Lay Navigators in Delivering Psychosocial Support to Older Adults With Cancer

Through semi-structured interviews, feedback will be qualitatively collected from community representatives to guide the adaptation and further refinement of lay navigator training materials.

The ability to train community lay navigators using the newly adapted training content- training sessions will be assessed by the number of training sessions completed by lay navigators and the research staff, allowing for sharing of broad perspectives on training methods, content, and delivery.

HDAC inhibitor (MK-0683)

The aim of the present study is to evaluate the efficacy and safety of MK-0683 in the treatment of PV and ET. This agent has most recently been shown to be a potent inhibitor of the autonomous proliferation of haematopoietic cells of PV and ET patients carrying the JAK2 V617F mutation. Accordingly, it may be anticipated that MK-0683 - by decreasing the JAK2 allele burden - may influence clonal myeloproliferation and in vivo granulocyte, platelet and endothelial activation , which are considered to be major determinants of morbidity and mortality ( thrombosis, bleeding, extramedullary haematopoiesis , myelofibrosis ) in these disorders. The effects of MK-0683 at the molecular level will be studied by global/ focused gene expression profiling, epigenome profiling and proteomics.
